CJC-1295 with DAC is a 29 amino acid growth hormone releasing hormone (GHRH) analogue bearing a Drug Affinity Complex — a maleimidopropionyl linker that forms a covalent bond with circulating serum albumin — identified by CAS number 863288-34-0. This albumin conjugation is the defining feature of the molecule: it shields the peptide from renal clearance and enzymatic degradation, extending the plasma half-life from roughly 30 minutes in the non-DAC form to approximately 8 days. Research has therefore studied the DAC variant in models of sustained growth hormone and IGF-1 elevation rather than the discrete pulses associated with Modified GRF (1-29), making the two forms distinct research tools rather than interchangeable ones. Like the non-DAC form it carries the D-Ala2, Gln8, Ala15 and Leu27 substitutions conferring dipeptidyl peptidase-4 resistance, and binds the GHRH receptor on anterior pituitary somatotrophs to initiate the G-protein coupled cascade through adenylyl cyclase, cAMP and protein kinase A.
